Everything discussed here is supplied strictly as a laboratory research material. None of these compounds is approved, intended or authorised for human consumption or therapeutic use by RS Bio Labs, and the trials and regulatory steps referenced describe other organisations' work. Orforglipron: an oral GLP-1 that is not a peptide
On 1 April 2026 the FDA approved orforglipron, marketed by Eli Lilly as Foundayo, for chronic weight management in adults with obesity, or with overweight plus a weight-related condition. According to coverage in Healio, the approval came under the agency’s National Priority Voucher pilot, roughly 50 days after filing, and the FDA described it as the fastest approval of a new molecular entity since 2002.
Two details matter to peptide researchers. First, orforglipron is a small molecule, not a peptide: it activates the GLP-1 receptor but is built to survive the gut, which is the problem peptide formulations have struggled with (see our look at the oral frontier). Second, it is taken once daily with or without food or water, unlike earlier oral semaglutide. Reported 72-week weight loss in the pivotal ATTAIN-1 programme was in the region of 11–12% depending on the analysis cited, with a label carrying the class thyroid-tumour warning. As always, compare figures only within the same estimand and trial design.
Survodutide: the glucagon/GLP-1 dual agonist reports
Survodutide is a dual GLP-1/glucagon receptor agonist developed by Boehringer Ingelheim with Zealand Pharma. Its first Phase 3 results arrived in 2026:
- SYNCHRONIZE-1 (obesity without diabetes): topline results in April 2026 reported weight loss of about 16.6% versus 3.2% on placebo at 76 weeks on the efficacy estimand. Secondary summaries of the ADA 2026 presentation quote a lower placebo-subtracted figure, so check the primary paper before quoting a single number.
- SYNCHRONIZE-MASLD (obesity with fatty liver disease): the co-primary endpoints were met, with mean body-weight change of −12.2% against −1.0% on placebo, and liver fat normalised in about six in ten participants.
- SYNCHRONIZE-2 (type 2 diabetes) is still to report.
Commentators have flagged a relatively high rate of discontinuation because of gastrointestinal side effects. The glucagon arm is the same design idea behind retatrutide’s triple agonism (see our TRIUMPH round-up): add liver and energy-expenditure pathways to the appetite effect. Survodutide’s liver-fat data are its most distinctive result.
The takeaway
2026 is shaping up as the year the field fans out: oral small molecules, dual and triple agonists, and combination approaches all reading out at once. For research purposes the mechanisms are what endure. Each design is a different answer to the same question — which receptors, in what ratio, with what delivery — and the trial data are beginning to put numbers against those choices. See the GLP-1 landscape map for how the approved and investigational compounds fit together.
